{"created":"2023-06-20T15:46:07.668470+00:00","id":695,"links":{},"metadata":{"_buckets":{"deposit":"7f48fc82-c9c3-4bb1-af40-0650fa52a8c5"},"_deposit":{"created_by":4,"id":"695","owners":[4],"pid":{"revision_id":0,"type":"depid","value":"695"},"status":"published"},"_oai":{"id":"oai:uoeh-u.repo.nii.ac.jp:00000695","sets":["13:25:28"]},"author_link":["1899","1906","1902","1905","1901","1898","1903","1900","1907","1904"],"item_10001_biblio_info_7":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2015-06-01","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"2","bibliographicPageEnd":"148","bibliographicPageStart":"139","bibliographicVolumeNumber":"37","bibliographic_titles":[{"bibliographic_title":"産業医科大学雑誌"},{"bibliographic_title":"Journal of UOEH","bibliographic_titleLang":"en"}]}]},"item_10001_description_5":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"アメリカ航空宇宙局による有人月面探査構想が発表され,我が国も参加を表明している.月面には,月レゴリスと呼ばれる微粒子を含む表土が堆積しており,月面での活動によって月レゴリスが舞い上がり,宇宙船内に持ち込まれて吸入する可能性がある.月面探査に先立ち,月レゴリスの有害性評価が必要であることから,本研究では月レゴリスの化学組成と粒径に着目し,細胞に対する影響を検討した.アポロが持ち帰った月レゴリスと同様の組成を持つ月レゴリスシミュラントを用い,液相沈降法により粒径が10 μm以下,25 μm以下および10~25 μmとなるように分画した.さらに,平均粒子径5.10 μmの微粒子からなる月レゴリスシミュラントを用い,0.1および1.0 mg/mlの濃度でヒト肺胞上皮腺癌由来のA549細胞およびマクロファージ様に分化させたヒト単球由来のTHP-1細胞に投与し,24時間後に,細胞傷害,酸化ストレス誘導および炎症誘導性を評価した.濃度1.0 mg/mlのときには,細胞膜の損傷とミトコンドリア活性の低下と,インターロイキン- 8( IL-8)の分泌および遺伝子発現の上昇が認められたが,0.1 mg/mlではこれらの影響はないか,ごく小さく,陽性対照とした結晶性SiO2 に比べても影響は小さかった.THP-1細胞において,濃度1.0 mg/mlでは培養上清中にIL-1βおよび腫瘍壊死因子-α( TNF-α)の分泌がみられたが,24時間の時点でIL-1βおよびTNF-α遺伝子の上昇はみられなかった.また,細胞内酸化ストレスの上昇も小さかった.細胞膜損傷,酸化ストレス誘発あるいは炎症誘発性といった細胞影響は<10 μmの画分で強い傾向があったが,顕著な違いはなかった.以上の結果から,in vitroの試験において化学組成の面からの細胞影響は小さく,粒子径の影響もそれほど大きくないことが示された.\nThe National Aeronautics and Space Administration has announced a plan to establish a manned colony on the surface of the moon, and our country, Japan, has declared its participation. The surface of the moon is covered with soil called lunar regolith, which includes fine particles. It is possible that humans will inhale lunar regolith if it is brought into the spaceship. Therefore, an evaluation of the pulmonary effects caused by lunar regolith is important for exploration of the moon. In the present study, we examine the cellular effects of lunar regolith simulant, whose components are similar to those of lunar regolith. We focused on the chemical component and particle size in particular. The regolith simulant was fractionated to < 10 μm, < 25 μm and 10-25 μm by gravitational sedimentation in suspensions. We also examined the cellular effects of fine regolith simulant whose primary particle size is 5.10 μm. These regolith simulants were applied to human lung carcinoma A549 cells at concentrations of 0.1 and 1.0 mg/ml. Cytotoxicity, oxidative stress and immune response were examined after 24 h exposure. Cell membrane damage, mitochondrial dysfunction and induction of Interleukin-8 (IL-8) were observed at the concentration of 1.0 mg/ml. The cellular effects of the regolith simulant at the concentration of 0.1 mg/ml were small, as compared with crystalline silica as a positive control. Secretion of IL-1β and tumor necrosis factor-α (TNF-α) was observed at the concentration of 1.0 mg/ml, but induction of gene expression was not observed at 24 h after exposure. Induction of cellular oxidative stress was small. Although the cellular effects tended to be stronger in the < 10 μm particles, there was no remarkable difference. These results suggest that the chemical components and particle size have little relationship to the cellular effects of lunar regolith simulant such as cell membrane damage, induction of oxidative stress and proinflammatory effect.
